avances en la embriología molecular y la evolución

Resumo

Nesta revisão, examinamos estudos de biologia molecular que revolucionaram os conceitos atuais sobre a evolução animal, dando origem à disciplina conhecida como Evo-Devo (biologia evolutiva do desenvolvimento). Primeiramente, discutimos a descoberta dos 39 genes Hox, que determinam a identidade corporal ao longo do eixo anteroposterior (A-P) tanto em vertebrados quanto em invertebrados. Em seguida, examinamos como um gradiente morfogenético determina o eixo dorsoventral (D-V) e a diferenciação
tecidual característica de todos os vertebrados. Esse gradiente forma-se durante a gastrulação por meio da secreção de Chordin, uma proteína que se liga às Proteínas Morfogenéticas Ósseas (BMPs) e as antagoniza; gera-se um fluxo através da degradação da Chordin pela proteinase Tolloid, a qual libera as BMPs em sua forma ativa no lado ventral. Analisamos como a atividade dessas redes gênicas — que regulam a comunicação celular embrionária — está na base de diversas patologias, como malformações congênitas e câncer. Confirmando o aforismo de que a oncogênese recapitula a ontogênese, o HoxA9 impulsiona a leucemia mieloide aguda; a β-catenina promove o câncer colorretal; a Nodal desencadeia a formação de células-tronco intratumorais; a Chordin encontra-se amplificada em tumores broncopulmonares; e a Goosecoid induz a transição epitélio-mesênquima, levando, assim, à metástase. Por fim, discutimos como essas descobertas modificaram os conceitos atuais sobre a evolução darwiniana por seleção natural. Nem todas as morfologias animais concebíveis eram possíveis; elas precisavam ser compatíveis com o uso de redes gênicas universais de desenvolvimento embrionário herdadas de Urbilateria, o ancestral comum de todos os animais bilaterais.
